应用高速逆流色谱法分离制备金银木叶中的芦丁,经过选择优化,确定以乙酸乙酯-正丁醇-水(5∶2∶5,v∶v)为两相溶剂系统,以上相为固定相,下相为流动相,在主机转速800 r/min,流速2 m L/min,检测波长256 nm条件下进行分离制备,得到4个分离组分。对所得分离产物进行高效液相检测,发现组分II为芦丁,其纯度达到96%以上,得率为10.7%。本实验从金银木叶中分离制备芦丁,方法简便,所得产物纯度高,为进一步扩大金银木的开发应用提供了理论依据。
Rutin was prepared by high-speed countercurrent chromatography. After optimization, it was confirmed that ethyl acetate-n-butanol-water (5:2:5, v:v) Phase and the lower phase as the mobile phase, and separated and prepared at a host speed of 800 r / min, a flow rate of 2 m L / min and a detection wavelength of 256 nm to obtain four separated components. The product was isolated by high performance liquid chromatography and found that component II is rutin, the purity of 96% or more, the yield was 10.7%. In this experiment, the isolation and preparation of rutin from the leaves of the honeysuckle, the method is simple, the purity of the resulting product is high, which provides a theoretical basis for further expanding the development and application of the honeysuckle.
