目的:分析百令胶囊对放射性肺损伤大鼠血浆IL-6、TNF-α、TGF-β1含量的影响,探讨百令胶囊防治放射性肺损伤效果及可能作用机制。方法:将141只6周龄SD大鼠(清洁级,雄性)随机分对照组(n=21)、模型组(n=30)、百令预防组(n=30)、百令治疗组(n=30)、地塞米松治疗组(n=30),各组大鼠均给予X射线全胸单次照射(20Gy),正常对照组除外。每组大鼠分别于第2、4、6周不同时间点随机处死,心脏采血约5mL,置肝素抗凝管中,采用ELISA法检测血浆中TNF-α、IL-6、TGF-β1含量。结果:相较于模型组,地塞米松组、百令预防组、治疗组大鼠第2、4、6周血浆中IL-6、TNF-α、TGF-β1含量显著下降(P<0.01或P<0.05);百令预防组大鼠第2、4、6周血浆中IL-6、TNF-α、TGF-β1含量低于地塞米松组和百令治疗组(P<0.01),百令治疗组大鼠第2、4、6周血浆中IL-6、TNF-α、TGF-β1含量低于地塞米松治疗组(P<0.01)。结论:百令胶囊可抑制或减轻放射性肺泡炎性反应、肺纤维化进程,可能通过降低血浆中IL-6、TNF-α、TGF-β1等细胞因子的含量实现。
Objective: To analyze the effect of Bailing capsule on the content of plasma IL-6, TNF-α and TGF-β1 in rats with radiation-induced lung injury and to explore the effect of Bailing capsule on radiation-induced lung injury and its possible mechanism. Methods: A total of 141 SD rats (clean grade, male) of 6 weeks old were randomly divided into control group (n = 21), model group (n = 30), Bailing prevention group n = 30) and dexamethasone treatment group (n = 30). All rats were given single X ray irradiation (20Gy), except for the normal control group. The rats in each group were randomly sacrificed at different time points at 2, 4, and 6 weeks. Blood samples were collected from 5 mL of blood and placed in heparin anticoagulant tubes. The levels of TNF-α, IL-6 and TGF-β1 were measured by ELISA. Results Compared with the model group, the levels of IL-6, TNF-α and TGF-β1 in the plasma of the dexamethasone group, the beraprost-preventive group and the treatment group were significantly decreased at the 2nd, 4th and 6th weeks (P <0.05). The levels of IL-6, TNF-α and TGF-β1 in the serum of rats in the Bailin prevention group were lower than those of the dexamethasone group and the Bailing group The levels of IL-6, TNF-α and TGF-β1 in the plasma of the treated group rats at 2, 4 and 6 weeks were lower than those of the dexamethasone treated group (P <0.01). Conclusion: Bailing Capsule can inhibit or reduce the radiation-induced alveolar inflammation, pulmonary fibrosis process may be through the plasma IL-6, TNF-α, TGF-β1 and other cytokines content achieved.
