目的研究热应激对大鼠睾丸诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)表达的影响及其意义。方法42d龄SD雄性健康大鼠12只建立单侧隐睾作为热应激处理的模型,术后第7d处死大鼠留取双侧睾丸,采用RT-PCR及免疫组化SABC法分别检测生精细胞iNOSmRNA和蛋白表达的变化,TUNEL法检测生精细胞凋亡。结果热应激处理后生精上皮内可见iNOS强阳性表达,而对侧睾丸生精上皮内无iNOS表达;热应激处理后睾丸iNOSmRNA也较对侧睾丸增强,生精细胞凋亡指数明显高于对侧睾丸,差异均有显著性意义(P<0.05)。结论热应激导致生精上皮内iNOS表达增强,生精细胞凋亡增加,iNOS参与介导生精细胞凋亡。
Objective To study the effect of heat stress on the expression of inducible nitric oxide synthase (iNOS) in rat testes and its significance. Methods Twelve male SD rats, aged 42d, were used to establish unilateral cryptorchidism model. The rats were sacrificed on the 7th day after surgery to take bilateral testes. RT-PCR and immunohistochemical SABC method were used to detect the spermatogenic protein The changes of iNOS mRNA and protein expression in the cells were detected by TUNEL method. Results The iNOS positive expression was observed in the seminiferous epithelium after heat stress, while the iNOS expression was not observed in the contralateral testis. The iNOS mRNA in the testes increased after heat stress treatment and the apoptosis index of spermatogenic cells was significantly higher than that in the contralateral testes Contralateral testis, the difference was significant (P <0.05). Conclusion Heat stress leads to enhanced expression of iNOS in spermatogenic epithelium and increased apoptosis of spermatogenic cells. INOS is involved in the apoptosis of spermatogenic cells.
