[目的]观察冠通方含药血清对体外培养的人外周血内皮祖细胞(EPCs)数量和功能的影响。[方法]将24只雄性Wistar大鼠随机分为冠通方组(低剂量组、中剂量组、高剂量组)和对照组,制备含药血清和空白血清。采用密度梯度离心法分离获得人外周血单个核细胞,加入各实验血清组培养液培养EPCs,经激光共聚焦显微镜、流式细胞仪鉴定为EPCs,分别在不同作用时间(12h,24h,48h,72h)后,倒置相差显微镜下观察并比较各组EPCs的数量、增殖能力、迁移能力和粘附能力。[结果]与对照组相比,冠通方组可促进外周血EPCs扩增,并显著改善外周血EPCs的粘附、迁移和增殖能力。[结论]冠通方可增加EPC的数量并改善其功能,这可能是冠通方防治冠心病患者经皮冠状动脉腔内成形术(PTCA)加支架植入术后再狭窄的作用机制之一。
[Objective] To observe the effect of Guan Tong Fang containing serum on the number and function of endothelial progenitor cells (EPCs) cultured in vitro. [Methods] Twenty-four male Wistar rats were randomly divided into Guan Tong Fang group (low dose group, medium dose group, high dose group) and control group, and drug-containing serum and blank serum were prepared. Peripheral blood mononuclear cells (PBMCs) were isolated by density gradient centrifugation. EPCs were cultured in culture medium of each experimental group. EPCs were identified by laser scanning confocal microscope and flow cytometry. The cells were cultured at different time (12h, 24h, 48h, 72h). The numbers of EPCs, proliferation, migration and adhesion were observed and compared under inverted phase contrast microscope. [Result] Compared with the control group, Guan Tong Fang group could promote EPCs expansion in peripheral blood and significantly improve the adhesion, migration and proliferation ability of EPCs in peripheral blood. [Conclusion] GuanTangFang can increase the number of EPCs and improve their function, which may be one of the mechanisms of GuanTongFang in the prevention and treatment of restenosis after percutaneous transluminal coronary angioplasty (PTCA) and stent implantation in patients with coronary heart disease .
