尽管发描电镜样品制备较透射电镜样品制备简单,但在样品制备过程中须用一些特殊的仪器,如临界点干燥仪、离子镀膜仪等。鉴于近年来微波辐射术在扫描电镜样品制备中的应用,本文采用微波辐射术对六胺银染色的扫描电镜样品(大鼠气管及十二指肠)进行了初步观察,取得了良好的结果。微波辐射是用SMC“一70E-2型”微波炉(广东顺德蚬华微波制品厂),输出功率650W,微波频率2450MHZ,火力档10档(PI-10)可调。样品经微波辐射固定(3%戊二醒)、后固定(1%OsO。)后,入l%过碘酸,辐射30秒,火力档PI;六胺银染色,辐射1分钟,火力档PZ。六胺银染液的配方:15%六次甲基四胺4ml,5%硼砂Zml,4%硼酸07ml,双蒸水25ml,染色前加入10%硝酸银lml,混匀。丙酮梯度脱水后,将样品置于微波炉内干燥,辐射2分钟,火力档P3,不需金属镀膜,即可在AMR-1000B型扫描电镜下观察。六胺银染色是通过过碘酸的氧化作用,将组织中的羧基氧化成醛基,而醛基能使六胺银还原成金属银,它均匀地沉积在含有凌基组织样品的表面,形成连续而均匀的金属薄膜,使样品获得良好的导电性能。微波干燥使经丙酮脱水的样品被整体加热,丙酮在组织内被迅速气化,使样品的干燥变得简单和快捷。本方法简便、快速,在制样过程无需特殊
