给440只Wistar大鼠喂饲含0、125、250、500及1000ppm富马酸二甲酯(DIMEFU)的饲料24个月。实验结果,各实验组与对照组相比,食物利用率、摄入量、生长、一般表现、死亡率及平均寿命均无显著性差异。血液学检查仅个别指标出现差别,但波动在正常范围。1000ppm组动物血清GPT、GOT及肝、肾脏器系数呈现持续性改变。病理组织学检查表明各组非肿瘤性及肿瘤性病变发生率与饲料中DIMEFU无关。大鼠长期摄入DIMEFU最大无作用剂量为250ppm,相当于25mg/kg。建议人的每日容许摄入量为0.25mg/kg。
440 Wistar rats were fed diets containing 0, 125, 250, 500 and 1000 ppm dimethyl fumarate (DIMEFU) for 24 months. As a result of the experiment, there was no significant difference in food utilization, intake, growth, general performance, mortality and average life span between the experimental groups and the control group. Hematological examinations differ only in individual indicators but fluctuate within the normal range. The serum GPT, GOT and hepatic and renal organ coefficients of the 1000ppm group showed persistent changes. Histopathological examination showed that the incidence of non-neoplastic and neoplastic lesions was not related to DIMEFU in feed. The maximum no-effect dose of the long-term intake of DIMEFU in rats was 250 ppm, equivalent to 25 mg/kg. The recommended daily intake of humans is 0.25 mg/kg.
