目的 :探讨骨肉瘤特异性细胞毒T淋巴细胞 (OSS CTL)对人骨肉瘤细胞系HOS 860 3细胞凋亡的诱导作用 ,以揭示其抗肿瘤机制。方法 :OSS CTL与HOS 860 3细胞按不同效靶比共同孵育后 ,通过电镜观察HOS 860 3细胞的超微结构变化 ,并用流式细胞仪定量检测OSS CTL与HOS 860 3细胞凋亡发生状况。结果 :HOS 860 3细胞被OSS CTL攻击后浓缩、深染 ,致密的染色质沿核膜下凝聚 ,并有凋亡小体形成。效靶比为 1∶1、5 0∶1、10 0∶1时 ,HOS 860 3细胞凋亡发生率分别为 0 .6%、12 .4%及 2 1.6%。结论 :OSS CTL可通过诱导人骨肉瘤细胞系HOS 860 3发生细胞凋亡起到抗肿瘤作用 ,且效靶比值增大后靶细胞凋亡的数量有明显增多的趋势
Objective: To investigate the induction of osteosarcoma-specific cytotoxic T lymphocytes (OSS CTL) on the apoptosis of human osteosarcoma cell line HOS 860 3 cells to reveal its anti-tumor mechanism. METHODS: OSS CTLs and HOS 860 3 cells were incubated with different ratios of target to target, and the ultrastructural changes of HOS 860 3 cells were observed by electron microscopy. Flow cytometry was used to quantitatively detect the apoptosis of OSS CTLs and HOS 860 3 cells. Results: HOS 860 3 cells were concentrated and deeply stained by OSS CTLs. Dense chromatin condensed along the nuclear membrane and formed apoptotic bodies. When the ratio of effect to target was 1:1, 50:1 and 10:0:1, the incidence of apoptosis in HOS 860 3 cells was 0.6%, 12.4%, and 21.6%, respectively. Conclusion : OSS CTL can induce anti-tumor effect by inducing apoptosis of human osteosarcoma cell line HOS 860 3 , and the number of target cell apoptosis increases after the ratio of effector to target is increased.
