目的探索重组人血清白蛋白-粒细胞集落刺激因子融合蛋白(GW003)对环磷酰胺(CTX)化疗大鼠的治疗作用。方法 80只SD大鼠经CTX化疗后分别一次皮下注射生理盐水和不同剂量GW003(250、500和1500μg/kg),每日一次检测外周血细胞计数,药后第10天处死大鼠取胸骨行病理组织学观察。结果 SD大鼠腹腔注射CTX后外周血白细胞、中性粒细胞、淋巴细胞和血小板数均显著下降。GW003治疗可明显升高化疗大鼠外周血白细胞和中性粒细胞数的最低值并缩短其减少的持续时间。GW003对CTX化疗大鼠外周血淋巴细胞、红细胞和血小板数无明显影响。结论 GW003对CTX化疗大鼠外周血白细胞减少有明显的治疗作用。
Objective To explore the therapeutic effect of recombinant human serum albumin-granulocyte colony-stimulating factor fusion protein (GW003) on the chemotherapy of cyclophosphamide (CTX) in rats. Methods Eighty SD rats were injected subcutaneously with normal saline and different doses of GW003 (250, 500 and 1500 μg / kg) respectively after the CTX chemotherapy. The peripheral blood cell counts were measured once a day. Histological observation. Results The number of leukocytes, neutrophils, lymphocytes and platelets in peripheral blood of SD rats decreased significantly after CTX injection. GW003 treatment can significantly increase the minimum number of peripheral blood leukocytes and neutrophils in chemotherapy rats and shorten the duration of their reduction. GW003 had no significant effect on the number of peripheral blood lymphocytes, erythrocytes and platelets in CTX chemotherapy rats. Conclusion GW003 has a significant therapeutic effect on CTX-induced peripheral blood leukopenia in rats.
