目的:观察4-羟苯维胺脂(4-HPR)联合阿霉素对人源性膀胱移行细胞癌细胞株T24的生长抑制和凋亡诱导作用,探讨两者是否存在协同作用。方法:采用MTT法评价4-HPR、阿霉素对T24细胞毒作用,应用流式细胞仪检测4-HPR、阿霉素或两者联合应用时对T24细胞生长抑制率。结果:4-HPR具有诱导T24细胞凋亡的作用,4-HPR与阿霉素联合用药有明显生长抑制和凋亡诱导的协同作用。结论:4-HPR对T24细胞的凋亡作用存在浓度和时间依赖性,与阿霉素联合用药能缩短抑瘤效果的作用时间,减少阿霉素的用量。
OBJECTIVE: To observe the effects of 4-HPR combined with doxorubicin on the growth inhibition and apoptosis induction of human bladder transitional cell carcinoma cell line T24 and to explore whether synergistic effects exist between them. Methods: MTT assay was used to evaluate the cytotoxicity of 4-HPR and doxorubicin to T24 cells. Flow cytometry was used to detect the inhibitory rate of 4-HPR, doxorubicin or a combination of the two on T24 cell growth. Results: 4-HPR induced the apoptosis of T24 cells. The combination of 4-HPR and doxorubicin significantly inhibited the growth and induced apoptosis. CONCLUSION: The apoptosis of T24 cells induced by 4-HPR is concentration-dependent and time-dependent. The combination of doxorubicin and doxorubicin can shorten the anti-tumor effect and reduce the dosage of doxorubicin.
