本研究旨在检测geminin和cdt1在初诊急性白血病(AL)患者外周血或骨髓细胞中的表达水平,并进一步探讨其在AL发病机制中的作用。采用SYBR Green实时定量逆转录聚合酶链反应(SYBR-RT-PCR)技术,检测初诊AL患者外周血或骨髓细胞geminin和cdt1mRNA的表达。结果表明:13例ALL初诊患者外周血中有10例孪蛋白(geminin)和cdt1mRNA表达阳性,阳性率为76.92%;14例AML初诊患者外周血中有9例表达阳性,阳性率为64.29%;而正常对照组中无阳性表达(0/10)。ALL和AML初诊患者骨髓细胞中geminin和cdt1mRNA表达(ALL:108.06±67.34,52.37±35.16;AML;62.66±58.69,26.68±22.29)均高于对照组(11.81±2.83,7.32±5.77),差异有统计学意义(p<0.01,p<0.05);34例初诊AL患者骨髓细胞中geminin和cdt1mRNA的表达呈明显的正相关(r=0.55,p<0.01)。结论 :geminin与cdt1在初诊AL患者中表达增高,且两者在AL骨髓细胞呈正相关,其过度表达可能在AL的发病机制中起重要作用。
The purpose of this study was to determine the expression of geminin and cdt1 in peripheral blood or myeloid cells of patients with newly diagnosed acute leukemia (AL) and to explore their roles in the pathogenesis of AL. SYBR Green Real-time quantitative reverse transcription-polymerase chain reaction (SYBR-RT-PCR) was used to detect the expression of geminin and cdt1mRNA in peripheral blood or bone marrow cells of newly diagnosed AL patients. The results showed that 10 cases of geminin and cdt1 mRNA were positive in the peripheral blood of 13 newly diagnosed ALL patients, the positive rate was 76.92%. In the 14 newly diagnosed AML cases, 9 cases were positive in peripheral blood, the positive rate was 64.29%. However, there was no positive expression in normal control group (0/10). The expression of geminin and cdt1mRNA in bone marrow cells of ALL and AML newly diagnosed patients (ALL: 108.06 ± 67.34,52.37 ± 35.16; AML; 62.66 ± 58.69,26.68 ± 22.29) were significantly higher than that of the control group (11.81 ± 2.83,7.32 ± 5.77) (P <0.01, p <0.05). The expression of geminin and cdt1 mRNA in 34 newly diagnosed AL patients was positively correlated (r = 0.55, p <0.01). CONCLUSION: The expressions of geminin and cdt1 in patients with newly diagnosed AL are increased, and both are positively correlated with AL bone marrow cells. The overexpression of geminin and cdt1 may play an important role in the pathogenesis of AL.
