通过单因素试验和正交试验探讨从黑皱鳃金龟中提取甲壳素的最佳工艺条件。利用氢氧化钠溶液和柠檬酸溶液依次对黑皱鳃金龟干粉进行处理后,用紫外吸收法和干法灰化法分别测定提取物中残余蛋白质和灰分含量以检验脱除效果,并在最佳工艺条件下,提取甲壳素样品,对其进行定性和定量分析。试验结果表明:利用酸碱法提取甲壳素的最佳工艺条件为:氢氧化钠浓度8%、固液比1∶10、温度85℃、时间4 h,柠檬酸浓度1.6 g/L、时间25 h、固液比1∶12、室温,对在此工艺条件下提取得到的甲壳素样品进行红外光谱检测并计算其得率为24.82%。
The optimum technological conditions of extracting chitin from Black-lipped beetles were studied by single factor test and orthogonal test. After using the sodium hydroxide solution and the citric acid solution to deal with the dry powder of Coprinus comatus in turn, the residual protein and ash content in the extract were measured respectively by UV absorption method and dry ashing method to test the removing effect, Chitin samples were extracted and analyzed qualitatively and quantitatively under the process conditions. The results showed that the optimum conditions for the extraction of chitin by acid-base method were: sodium hydroxide concentration 8%, solid-liquid ratio 1:10, temperature 85 ℃, time 4 h, citric acid concentration 1.6 g / L, time 25 h, solid-to-liquid ratio 1:12, room temperature. The chitin samples extracted under the above process conditions were detected by infrared spectrum and the yield was 24.82%.
