目的 :研究 P物质 (SP)在不同水平对大鼠黄体生成素 (LH)和睾酮 (T)分泌的影响。方法 :下丘脑、腺垂体、睾丸离体组织培养以及顺序灌注 ,于三级组织培养液中加入不同浓度的 SP,用放射免疫分析法测定培养液中 LH和 T的分泌量。结果 :不同浓度的 SP在睾丸水平均可直接抑制 T的分泌 (P<0 .0 1 ) ,而在腺垂体水平对 L H的分泌则无显著影响 (P>0 .0 5) ;经顺序灌注后SP(50 0 0 ng/ 1 0 ul)可在下丘脑水平间接抑制 LH和 T的分泌 (P<0 .0 1 )。结论 :SP可能作为一种抑制性调节肽参与了生殖内分泌活动的调节。
Objective: To study the effects of substance P (SP) on the secretion of luteinizing hormone (LH) and testosterone (T) in rats. Methods: Hypothalamus, adenohypophyseal and testis were cultured in vitro and sequential perfusion. Different concentrations of SP were added into the three tissue culture media. The secretion of LH and T in the culture media was determined by radioimmunoassay. Results: Different concentrations of SP could directly inhibit the secretion of T (P <0.01) at the testicular level, but had no significant effect on the secretion of LH at the pituitary gland (P> 0.05). After sequential perfusion Post-SP (500 ng / 100 ul) could indirectly inhibit LH and T secretion at the hypothalamus level (P <0.01). Conclusion: SP may participate in the regulation of reproductive and endocrine activities as an inhibitory regulatory peptide.
