氯芬待因是由双氯芬酸和磷酸可待因组成的复方动物长期灌胃氯芬待因后,10mg·kg-1·d-1组大鼠有9/20只死亡,5mg·kg-1·d-1组大鼠有1/20只死亡,2mg·kg-1·d-1组大鼠未出现死亡。6.4mg·kg-1·d-1组狗有2/4只死亡,2mg·kg-1·d-1组狗未出现死亡。除2mg·kg-1·d-1组大鼠的体重和食量变化不明显外,给药组大鼠和狗的食量和体重都有明显减少(P<0.05)。外观症状主要表现为:拉稀,黑便及便血,四肢无力,走路左右摇摆,有的甚至卧地不起,个别动物因衰竭而死亡。血液生化测定表明,10mg·kg-1·d-1组大鼠的血液总胆固醇含量增高(P<0.05),总蛋白量下降(P<0.05),6.4mg·kg-1·d-1组狗的总胆固醇虽也有增高趋势,但无统计意义。病理学损伤主要在肝、肾和消化道,肝组织充血,肝细胞混浊肿胀、空泡及坏死,肾小管细胞浊肿及核消失,肠胀气,部分胃、肠道粘膜脱落坏死,个别严重者出现胸水和腹水。血液学、尿常规、心电图及脏器系数测定结果表明给药组和对照组没有显著性差异。恢复期大鼠和狗的食量和体重都有明显回升,血液生化指标和病理变化也都有好转,说明该药对动物所?
