目的 :探讨膀胱移行细胞癌核基质蛋白 (nuclearmatrixproteins ,NMPs)与膀胱移行细胞癌的相关性。方法 :根据Wen等的方法改良制备膀胱移行细胞癌NMPs ;NMPs免疫家兔获得抗血清 ,利用免疫吸附法及硫酸铵沉淀法纯化抗血清 ;Westernblot及confocal分析膀胱移行细胞癌NMPs与其他对照组之间的差异性。结果 :膀胱癌NMPs具有 (2 0~ 4 0 )× 10 3 及 (6 0~ 90 )× 10 3 两富集蛋白条带区域 ,与对照组比较差异明显 ;膀胱移行细胞癌中NMPs主要表达于细胞核、核膜及部分胞质 ,而对照组织无明显特异性表达 ,差异有显著意义。结论 :膀胱移行细胞癌存在肿瘤相关的特异性NMPs ,为开发基于尿液或血液的免疫检测方法提供了实验基础。
Objective: To investigate the relationship between nuclear matrix proteins (NMPs) and transitional cell carcinoma of the bladder. Methods: The bladder transitional cell carcinoma (NMPs) were prepared according to the method of Wen et al. The antiserum was obtained from the rabbits immunized with NMPs. The antiserum was purified by immunosorbent assay and ammonium sulfate precipitation. The expressions of NMPs in bladder transitional cell carcinoma and other control groups were analyzed by Western blot and confocal analysis Differences between. Results: The NMPs in bladder transitional cell carcinoma showed the bands of (20 ~ 40) × 10 3 and (60 ~ 90) × 10 3, which were significantly different from the control group. The expression of NMPs in bladder transitional cell carcinoma Nuclear, nuclear membrane and some cytoplasm, while the control tissue no specific expression, the difference was significant. CONCLUSIONS: Tumor-associated specific NMPs are present in transitional cell carcinoma of the bladder and provide an experimental basis for the development of a urine or blood-based immunoassay.
