从溶液中回收或精制胰岛素最常用的方法是结晶法。一般是采用锌盐结晶法,而胰岛素钠盐结晶的制备国内还未见报导。据Jackson报导,可在胰腺酸醇粗提液中直接采用这种结晶法,收率每公斤胰腺可达2600~4600IU。我们利用层析法去除二价离子后的猪胰岛素溶液,进行钠盐结晶制备的探索。实验部分基本步骤:去二价阳离子猪胰岛素溶液加NaOH调pH为7.2~10.0,搅拌下加入NaCl使溶液Na~+总浓度在0.2~1.0M范围内,室温下机械搅拌0.25~72小时,分离结晶体,母液回收重复上述步骤。
