目的探讨半枝莲提取物(ESB)对人肝癌Hep-G2细胞体外增殖的影响及其作用机制。方法以MTT法检测ESB对Hep-G2细胞增殖的影响;光镜、电镜观察ESB作用后Hep-G2细胞形态的变化;流式细胞仪检测ESB作用后Hep-G2凋亡细胞、细胞周期的变化及凋亡相关蛋白Bcl-2,Bax,Fas表达。结果ESB能明显抑制Hep-G2细胞的增殖,并有剂量效应及时间效应关系。ESB作用Hep-G2细胞72h后,呈现凋亡早期的形态学改变;凋亡细胞增多;S期细胞减少,G0/G1期细胞增多;Fas表达增强,Bcl-2及Bax表达变化不明显。结论ESB可抑制Hep-G2细胞增殖,阻滞细胞周期,促进细胞凋亡,可能与激活FNFR超家族有关。
Objective To investigate the effects of Scutellaria barbata extract (ESB) on the proliferation of Hep-G2 cells in vitro and its mechanism. Methods The effect of ESB on the proliferation of Hep-G2 cells was detected by MTT assay. Morphological changes of Hep-G2 cells were observed under light microscope and electron microscope. The apoptosis of Hep-G2 cells were detected by flow cytometry And apoptosis related protein Bcl-2, Bax, Fas expression. Results ESB could significantly inhibit the proliferation of Hep-G2 cells with dose-response and time-effect. After treated with ESB for 72 hours, the morphological changes of apoptotic cells were observed. The number of apoptotic cells increased, the number of S phase cells decreased, the number of G0 / G1 phase cells increased, the expression of Fas increased but the expression of Bcl-2 and Bax did not change obviously. Conclusion ESB can inhibit Hep-G2 cell proliferation, arrest cell cycle and promote apoptosis, which may be related to activation of FNFR superfamily.
