目的探讨大豆异黄酮总提取物对荷瘤去卵巢鼠雌激素依赖型乳腺肿瘤的影响。方法以6.25,12.5和25 g/kg bw剂量大豆异黄酮总提取物喂食裸鼠2 w,去除卵巢并种植MCF-7肿瘤细胞,继续喂食16 w。用常规免疫组织化学方法检测肿瘤细胞核抗原Ki-67的表达,实时定量PCR(real-time PCR)检测雌激素相关基因pS2表达,化学发光酶免疫法检测裸鼠血清中雌激素的含量。结果与对照组比,喂食大豆异黄酮总提取物组裸鼠MCF-7移植瘤体积明显增加(P<0.05),肿瘤细胞增殖能力显著增强(P<0.05),pS2基因的表达增强(P<0.05),同时血清中雌激素的水平也因喂食大豆异黄酮总提取物而升高(P<0.05)。结论在本实验浓度范围内,青春期喂食大豆异黄酮总提取物对去卵巢裸鼠雌激素依赖型乳腺癌细胞移植肿瘤的生长有促进作用。
Objective To investigate the effects of total soy isoflavones extract on estrogen-dependent breast tumors in tumor-bearing ovariectomized mice. Methods The total extract of soybean isoflavones at doses of 6.25, 12.5 and 25 g / kg bw were injected into nude mice for 2 weeks. The ovaries were removed and MCF-7 tumor cells were planted. The rats were fed for another 16 weeks. The expression of Ki-67 was detected by routine immunohistochemistry. The expression of estrogen-related gene pS2 was detected by real-time PCR. The level of estrogen in nude mice was detected by chemiluminescence enzyme immunoassay. Results Compared with the control group, the volume of MCF-7 xenografted in total extract of soybean was significantly increased (P <0.05), the proliferation of tumor cells significantly increased (P <0.05) and the expression of pS2 gene enhanced (P < 0.05). Meanwhile, the level of estrogen in serum also increased due to the total extract of soy isoflavones (P <0.05). Conclusion In this experimental concentration range, adolescent fed soy isoflavones total extract on ovariectomized nude mice estrogen-dependent breast cancer cell transplantation tumor growth promotion.
