目的 研究脱氢异雄酮 (DHEA)对高糖下视网膜毛细血管周细胞增殖的影响。方法 以新鲜牛眼球为材料 ,分离并培养周细胞 ;采用 3H- Td R掺入法及液体闪烁技术 ,研究不同浓度的 DHEA(10、5 0、10 0、5 0 0、10 0 0 nmol/L)对周细胞增殖的影响。结果 高浓度葡萄糖 (2 0 mmol/L)培养液中加入 DHEA(5 0、10 0、5 0 0、10 0 0 nmol/L)培养周细胞 48小时 ,周细胞的增殖与对照组比较均有显著性差异 ,其中 DHEA为 5 0 0 nmol/L时对周细胞增殖的促进作用最大 (增殖率为42 .6 7% )。结论 DHEA可促进高糖下周细胞的增殖 ,这为筛选临床防治糖尿病视网膜病变的有效药物提供了可靠的依据。
Objective To study the effect of dehydroepiandrosterone (DHEA) on the proliferation of retinal capillary pericytes under high glucose. Methods Fresh bovine eyeballs were used as materials to separate and culture pericytes. 3H-TdR incorporation and liquid scintigraphy were used to study the effects of different concentrations of DHEA (10,5 0,10 0,5 0,10 0 0 nmol / L) on the proliferation of peripheral blood cells. RESULTS: Peripheral blood cells were cultured in high glucose (20 mmol / L) for 48 hours after adding DHEA (50, 100, 500, 10000 nmol / L). The proliferation of pericytes compared with the control group There was significant difference between the two groups. DHEA of 500 nmol / L could promote the proliferation of pericytes (the proliferation rate was 42.67%). Conclusions DHEA can promote the proliferation of the next week cells under high glucose, which provides a reliable basis for the screening of effective drugs for the prevention and treatment of diabetic retinopathy.
