测定血、尿等生物样品中阿洛西林(Azl-ocillin,苯咪唑青霉素)含量的高效液相色谱法已有报道。但其有关杂质的分离检出方法甚少见,仅有采用梯度洗脱系统分离其噻唑酸(Azlocilloic acid,Ⅱ) 的介绍。为对阿洛西林钠(Ⅰ)和有关主要杂质包括(Ⅱ)、侧链酸α-(α-氧-1-咪唑烷基甲酰胺基)苯乙酸(IPA)以及 6-氨基青霉烷酸(6APA)进行分离和定量,我们在YWG-C_(18)柱上,就甲醇。磷酸盐缓冲液溶剂系统进行了一些筛选。在选定的色谱条件下,除可对(Ⅰ)进行定量分析外,还能用于上述杂质的定性检别及其含量的限度检查。
Determination of blood, urine and other biological samples of azlocillin (Azl-ocillin, benzimidazole) content of high performance liquid chromatography has been reported. However, the methods for the separation and detection of impurities are very rare. Only the introduction of gradient elution system for the separation of azlocilloic acid (Ⅱ) has been reported. To investigate the effect of azlocillin sodium (I) and related major impurities including (II), the side chain acid alpha- (alpha-oxo- 1 -imidazolidinylcarboxamido) phenylacetic acid (IPA) and 6-aminopenicillanic acid (6APA) for separation and quantification, we methanol on YWG-C_ (18) column. Phosphate buffer solvent systems were screened. In the selected chromatographic conditions, in addition to (Ⅰ) for quantitative analysis, but also for the qualitative detection of impurities and limit the content of inspection.
