目的:建立测定人血浆中氢氯噻嗪浓度的RP-HPLC法,并用于研究氢氯噻嗪在人体的代谢过程。方法:血浆经甲基叔丁基醚提取后,以Shimadzu Shim-pack VP-ODS(5μm,150mm×4.6mm)为色谱柱;流动相为磷酸四甲基铵缓冲水溶液(pH=4.90)-乙腈(85∶15);流速1.0mL·min-1;检测波长271nm。结果:氢氯噻嗪在2.0~400ng·mL-1浓度范围内线性关系良好(r=0.9991);测定方法回收率在94.1%~109.0%之间,日内RSD<5%,日间RSD<10%。结论:本文所建立的方法灵敏度好、准确率高,可用于氢氯噻嗪的药代动力学研究和临床药物浓度的监测。
Objective: To establish a RP-HPLC method for the determination of hydrochlorothiazide in human plasma and to study the metabolic process of hydrochlorothiazide in the human body. Methods: The plasma was extracted with methyl tert-butyl ether (TMP) and the column was Shimadzu Shim-pack VP-ODS (5μm, 150mm × 4.6mm). The mobile phase consisted of tetramethylammonium phosphate buffered aqueous solution (85:15); flow rate 1.0mL · min-1; detection wavelength 271nm. Results: The linear range of hydrochlorothiazide was 2.0-400ng · mL-1 (r = 0.9991). The recoveries were 94.1% -109.0%. The RSDs were less than 5% and the daytime RSD was less than 10%. Conclusion: The method established in this paper has good sensitivity and high accuracy, which can be used to study the pharmacokinetics of hydrochlorothiazide and the monitoring of clinical drug concentration.
