目的:探讨甲基莲心碱(neferine)对氧化损伤的大鼠乳鼠心肌细胞的保护作用。方法:分离培养SD乳鼠心肌细胞,建立心肌细胞过氧化损伤模型,将细胞分为7组,即空白对照组(control)、溶剂对照组(DMSO)、氧化损伤组(H2O2)、氧化损伤加入槲皮素对照组(quercetin+H2O2)、氧化损伤加入莲心碱低、中、高浓度组(甲基莲心碱+H2O2、甲基莲心碱-M+H2O2、甲基莲心碱-H+H2O2)。将300μmol/LH2O2作用于加入槲皮素及不同浓度甲基莲心碱预培2 h的心肌细胞,继续培养24h,以细胞毒四唑盐(MTT)比色试验,乳酸脱氢酶(LDH)、谷胱甘肽过氧化物酶(GSH-px)、丙二醛(MDA)、流式细胞凋亡率、Caspase酶活性为检测指标。结果:甲基莲心碱呈剂量依赖性降低H2O2对心肌细胞活力的影响,降低MDA、LDH量,增加细胞GSH-px活性,并显著抑制Caspase-3和9活性,减少细胞凋亡数量,各指标差异具有统计学意义(P<0.01)。结论:甲基莲心碱可保护和修复过氧化氢诱导的心肌细胞的损伤,其作用可能是通过抗氧化、增加细胞GSH-px活性,进而抑制Caspase-3和9活性,减少细胞凋亡。
Objective: To investigate the protective effects of neferine on oxidative damage of rat neonatal rat cardiomyocytes. Methods: Myocardial cells were isolated from SD neonatal rats and the model of peroxisome cell injury was established. The cells were divided into 7 groups: blank control group, solvent control group (DMSO), oxidative damage group (H2O2) Quercetin + H2O2, and oxidative damage, low, medium and high concentration of neferine (norepinephrine + H2O2, neferine-M + H2O2, neferine-H + H2O2) 300μmol / LH2O2 was added to the myocardial cells pretreated with quercetin and different concentrations of neferine for 2 h, and the cells were cultured for 24 h. MTT colorimetric assay, lactate dehydrogenase (LDH) Glutathione peroxidase (GSH-px), malondialdehyde (MDA), flow cytometry and Caspase activity were detected. Results: Neferine decreased the effect of H2O2 in a dose-dependent manner on the viability of cardiomyocytes, decreased the content of MDA and LDH, increased the activity of GSH-px, significantly inhibited the activity of Caspase-3 and 9 and the number of apoptotic cells. The difference was statistically significant (P <0.01). CONCLUSION: Neferine protects and repairs hydrogen peroxide-induced injury of cardiomyocytes. Its action may be through anti-oxidant, increasing the activity of GSH-px, thereby inhibiting the activity of Caspase-3 and 9 and decreasing the apoptosis of cardiomyocytes.
