目的 用反义核糖核酸 (RNA)封闭尿激酶受体的表达 ,抑制人肺癌细胞株 95D的侵袭rrrrrrrrrn转移能力 ,验证反义RNA技术在抗肿瘤中的作用。方法 将尿激酶受体 (uPAR)反义核糖核酸表达质rrrrrrrrrn粒转染具有高度侵袭转移能力的人肺癌细胞株 95D ,利用改良Boyden小室分析转染细胞的体外侵袭rrrrrrrrrn能力 ,并将转染细胞接种裸小鼠以观察其体内转移能力。结果 G41 8筛选后 ,鉴定出 2个表达uPARrrrrrrrrrn反义RNA细胞克隆 ,uPAR蛋白质水平相应降低。表达uPAR反义RNA的细胞克隆的体外侵袭能力rrrrrrrrrn及在裸小鼠体内的肺转移能力较亲本细胞和空载体对照细胞均有显著性降低 (P < 0 .0 5)。结论 抑rrrrrrrrrn制尿激酶受体的表达 ,能够有效抑制肺癌的侵袭转移 ,反义RNA技术在抗肿瘤治疗中有良好的应用rrrrrrrrrn前景。
Objective To use antisense ribonucleic acid (RNA) to block the expression of urokinase receptor and inhibit the invasion of human lung cancer cell line 95D.rrrrrrrrrnThe ability to metastasize was verified. METHODS: Human urokinase receptor (uPAR) antisense RNA expression plasmids were transfected into human lung cancer cell line 95D with high invasion and metastasis capacity. The in vitro invasiveness of the transfected cells was analyzed using a modified Boyden chamber and the transfected cells were inoculated with nude mice to observe their ability to metastasize in vivo. Results After G41 8 screening, 2 antisense RNA cell clones expressing uPARrrrrrrrrrn were identified, and the uPAR protein level was correspondingly decreased. In vitro invasiveness of cell clones expressing uPAR antisense RNArrrrrrrrrn and lung metastasis capacity in nude mice compared to parental cells and empty vector control cells Significantly decreased (P < 0.05). Conclusion Inhibition of urokinase receptor expression can effectively inhibit the invasion and metastasis of lung cancer. Antisense RNA technology is good in anti-tumor therapy ApplyrrrrrrrrrnForeground.
