目的:观察蛴螬提取物对人肺癌A549细胞增殖的影响及其机制。方法:采用MTT法检测蛴螬提取物对人肺癌A549细胞的生长抑制率;用HE染色方法观测凋亡细胞的形态学变化;运用免疫组织化学SP法检测用药前后细胞周期蛋白D1(CyclinD1)的表达改变。结果:用蛴螬提取物处理的人肺癌A549细胞生长抑制率明显高于对照组;施药早期细胞出现凋亡形态学变化;A549细胞经蛴螬提取物处理后CyclinD1表达均下降。结论:蛴螬提取物在体外对人肺癌A549细胞株有显著的增殖抑制作用,其机制与下调CyclinD1的表达有关。
OBJECTIVE: To observe the effect and its mechanism of the extract of Lycium barbarum on the proliferation of human lung cancer A549 cells. METHODS: MTT assay was used to detect the growth inhibition rate of Lycium barbarum extract on human lung cancer A549 cells; HE staining was used to observe the morphological changes of apoptotic cells; SP immunohistochemical method was used to detect the expression of cyclin D1 before and after treatment. change. RESULTS: The growth inhibitory rate of human lung cancer A549 cells treated with wolfberry extract was significantly higher than that of the control group. Apoptotic morphological changes appeared in the early stage of application of the cells, and the expression of CyclinD1 was decreased in the A549 cells treated with wolfberry extract. Conclusion: Scorpion extract has a significant inhibitory effect on human lung cancer A549 cell line in vitro, and its mechanism is related to the down-regulation of CyclinD1 expression.
