目的:研究超临界CO2流体萃取南方红豆杉叶中脂肪油的最佳工艺及其成分分析。方法:以南方红豆杉叶脂肪油萃取得率为评价指标,考察不同萃取压力、萃取温度、萃取时间对脂肪油萃取率的影响,并采用衍生化GC-MS法对南方红豆杉叶脂肪油进行成分分析。结果:超临界CO2萃取南方红豆杉叶脂肪油最终优化的萃取工艺为:CO2压缩泵频率为10 Hz,萃取压力为25 MPa,萃取温度为45℃,萃取时间为120 min,分离釜Ⅰ压力8.0 MPa、温度40℃,分离釜Ⅱ压力5.0 MPa、温度35℃。萃取得到的脂肪油经三氟化硼衍生化后,经GC-MS分析,共鉴定出了33种脂肪酸。结论:3种不同产地的南方红豆杉叶脂肪油的萃取率不同,且以无锡东港的萃取率最高为2.61%。对其成分的分析,不同产地和不同提取方法下主要脂肪酸种类基本一致,含量较低的脂肪酸种类差别较大。
OBJECTIVE: To study the optimum technology and composition analysis of fatty oil extraction from S. yew leaf by supercritical CO2 fluid. Methods: The extraction yield of fatty oil from southern yew leaf was used as evaluation index to investigate the effects of different extraction pressure, extraction temperature and extraction time on the extraction rate of fatty oil. The derivatization GC-MS Component Analysis. Results: The optimum extraction conditions of supercritical CO2 extraction of fatty oil of Taxus cuspidata leaves were as follows: the CO2 compression pump frequency was 10 Hz, the extraction pressure was 25 MPa, the extraction temperature was 45 ℃, the extraction time was 120 min, the separation kettle pressure was 8.0 MPa, temperature 40 ℃, pressure of separation kettle 5.0 MPa, temperature 35 ℃. Fatty oil extracted by boron trifluoride derivatization, GC-MS analysis, a total of 33 kinds of fatty acids identified. CONCLUSION: The extraction rates of the fatty oils from Taxus cuspidata leaves in three different habitats are different, and the highest extraction rate in Donggang Wuxi is 2.61%. The analysis of its components, different origin and different extraction methods are basically the same type of fatty acids, fatty acids with lower levels of different species.
