目的研究氢氟酸烧伤中毒后对兔外周血单个核细胞早期凋亡百分率和胞内游离钙浓度([Ca~(++)]i)的影响。方法 用流式细胞仪检测兔烧伤染毒前后外周血单个核细胞的Annexin V变化,以观察其早期凋亡百分率;用Fluo-3/Am荧光探针观察烧伤染毒前后外周血单个核细胞内Ca~(++)平均荧光强度值的变化。结果 12只烧伤中毒兔外周血单个核细胞早期凋亡百分率显著增加,较染毒前比较P<0.01。而12只烧伤中毒兔染毒后1h外周血单个核细胞内[Ca~(++)]i与染毒前比较P>0.05,差异无显著性。结论 本实验提示氯氟酸烧伤中毒引导的细胞凋亡不同于一般认为钙离子浓度升高是大多数细胞凋亡所必需的途径,而是别的凋亡机制。
Objective To investigate the effect of hydrofluoric acid burn on the percentage of early apoptosis and the intracellular free calcium concentration ([Ca ~ (++)] i in peripheral blood mononuclear cells of rabbits. Methods The changes of Annexin V in peripheral blood mononuclear cells before and after burn in rabbits were detected by flow cytometry to observe the percentage of early apoptosis. Fluo-3 / Am fluorescence probe was used to observe the changes of the expression of Annexin V in peripheral blood mononuclear cells Changes of mean fluorescence intensity of Ca ~ (++). Results The percentage of early apoptotic cells in peripheral blood mononuclear cells of 12 poisoning rabbits increased significantly compared with those before exposure (P <0.01). However, there was no significant difference in [Ca ~ (++)] i in peripheral blood mononuclear cells at 1 hour after exposure to 12 burned poisoned rabbits. Conclusions This study suggests that apoptosis induced by chlorofluoric acid burn poisoning is different from that generally believed that the increase of calcium ion concentration is a necessary pathway for the majority of apoptosis, but another apoptosis mechanism.
