目的研究缺氧对大鼠嗜铬细胞瘤(PC12)细胞中白细胞介素6(IL-6)和IL-6受体β亚基(GP130)表达的影响。方法制备缺氧细胞模型。采用放射免疫技术、逆转录-聚合酶链反应(RT-PCR)技术和免疫蛋白印迹(W estern B lot)法检测IL-6基因mRNA和活性、GP130基因mRNA和蛋白表达水平。结果PC12细胞在缺氧后2 hIL-6mRNA(1.121±0.076)和活性〔(167.27±12.03)pg/m l〕、GP130mRNA(1.695±0.089)和蛋白表达(1.652±0.137)增强(P<0.05),6 h〔分别为(1.431±0.115),(320.66±11.82)pg/m l,(2.134±0.141),(2.521±0.164)〕达到高峰(P<0.01),显著高于对照组〔分别为(0.897±0.073,69.71±8.51)pg/m l,(1.356±0.082),(1.029±0.032)〕。结论缺氧能增强IL-6和GP130基因的表达。
Objective To investigate the effects of hypoxia on the expression of interleukin 6 (IL-6) and IL-6 receptor β subunit (GP130) in rat pheochromocytoma (PC12) cells. Methods The hypoxic cell model was prepared. The mRNA and activity of IL-6 gene and the expression of GP130 mRNA and protein were detected by radioimmunoassay, reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot. Results PC12 cells were enhanced (P <0.05) at 1 h after hypoxia (1.121 ± 0.076) and activity [(167.27 ± 12.03) pg / ml〕, GP130 mRNA (1.695 ± 0.089) and protein expression (1.652 ± 0.137) 6h 〔(1.431 ± 0.115), (320.66 ± 11.82〕 pg / ml, (2.134 ± 0.141), (2.521 ± 0.164〕〕 (P <0.01), which were significantly higher than that of the control group ± 0.073, 69.71 ± 8.51) pg / ml, (1.356 ± 0.082), (1.029 ± 0.032)]. Conclusion Hypoxia can enhance the expression of IL-6 and GP130 genes.
