目的:研究大孔树脂纯化二仙汤有效部位群的最佳条件。方法:以复方二仙汤中总黄酮、总生物碱、总皂苷为指标,以动态法考察D101,DM130,HPD100,HPD400 4种树脂对各指标的吸附和解析性能,优选出适宜的树脂,并进一步优化其吸附与解析条件。结果:D101型树脂较适宜纯化复方二仙汤有效部位群,最佳条件为二仙汤调药液比1∶1.5,加NaCl调至1.5mol.L-1,以2.5 BV上样,6 BV水洗至近无色,再用5 BV的50%乙醇洗脱,得有效部位群。结论:D101型树脂纯化复方二仙汤有效部位群工艺简单可行。
Objective: To study the optimal conditions for the purification of effective fractions of Erzhu Decoction by macroporous resin. Methods: The total flavonoids, total alkaloids and total saponins of Fufang Erxian decoction were used as indexes to investigate the adsorption and analytical properties of D101, DM130, HPD100 and HPD400 by dynamic method. Further optimization of its adsorption and analysis conditions. Results: The D101 resin was more suitable for the purification of the active fractions of Fufang Erxian Decoction. The optimum conditions were as follows: the ratio of Erxian Decoction to the solution was 1: 1.5, NaCl was added to 1.5 mol·L-1, Washed nearly colorless, and then 5 BV of 50% ethanol elution, the effective part of the group. Conclusion: The D101-type resin is effective and feasible for the purification of the effective fractions of Fufang Erxian Decoction.
