目的:建立用超高效液相色谱(UPLC)仪与蒸发光散射检测器联用同时测定北柴胡中柴胡皂苷a,c,d含量的方法。方法:采用UPLC仪,BEH C18色谱柱(2.1 mm×50 mm,1.7μm),流动相乙腈-水,梯度洗脱,流速0.4 mL.min-1,进样量3μL,柱温30℃,样品管理器温度4℃,漂移管温度40℃,氮气流速2.5 L.min-1。结果:可在6 min内完成1次色谱分析,柴胡皂苷a,c,d色谱峰之间有良好的分离度,柴胡皂苷a,c,d分别在0.043 6~0.348 8,0.046 8~0.374 4,0.049 2~0.393 6μg线性关系良好,精密度、重复性及加样回收率的RSD均<3.0%。结论:方法快捷、准确,重复性好,能同时测定柴胡皂苷a,c,d的含量。
Objective: To establish a method for the simultaneous determination of a, c and d of saikosaponin in Bupleurum chinense by ultra performance liquid chromatography (UPLC) and evaporative light scattering detector. Methods: The mobile phase of acetonitrile-water was eluted with a BEH C18 column (2.1 mm × 50 mm, 1.7 μm) using a UPLC instrument at a flow rate of 0.4 mL · min-1 with a sample volume of 3 μL and a column temperature of 30 ° C. Manager temperature 4 ℃, drift tube temperature 40 ℃, nitrogen flow rate 2.5 L.min-1. Results: The chromatographic analysis was completed within 6 min. The separation of saikosaponin a, c and d peaks was good. The concentrations of saikosaponin a, c and d were 0.043 6 ~ 0.348 8 and 0.046 8 ~ 0.374 4,0.049 2 ~ 0.393 6μg good linear relationship, precision, repeatability and sample recovery RSD were <3.0%. Conclusion: The method is rapid, accurate and reproducible. The contents of a, c and d of saikosaponin can be determined simultaneously.
