目的:研究清热化痰法对气道黏液高分泌大鼠气道炎症的影响及机制。方法:将48只SD大鼠随机分为6组,每组8只,分别为空白组、模型组、低剂量止咳颗粒组、中剂量止咳颗粒组、高剂量止咳颗粒组、对照组,采取BALF细胞计数及分类观察炎症细胞、HE染色观察肺组织病理变化及ELISA检测血清NE等方法。结果:①细胞计数及分类:空白组、中、高剂量止咳颗粒组中性粒细胞、嗜酸性粒细胞、淋巴细胞较模型组、低剂量止咳颗粒组低(P<0.05或P<0.01)。②HE染色:正常大鼠气道黏液较少;模型组大鼠炎性细胞浸润及分泌物阻塞气道,肺泡壁多处断裂,部分肺泡融合成肺大泡;低剂量止咳颗粒组较模型组无明显变化;中剂量止咳颗粒组气管、支气管炎症细胞浸润减少;高剂量止咳颗粒组支气管管壁分泌物堵塞少,上皮细胞排列尚整齐;对照组肺泡壁断少,偶可见少许炎细胞。③血清NE:空白组、高剂量止咳颗粒组血清NE含量较模型组、低、中剂量组止咳颗粒组低(P<0.01)。结论:杏杷止咳颗粒能够通过清热化痰法减轻气道黏液高分泌大鼠气道炎症,其机制可能与血NE下降有关。
Objective: To study the effect of clearing heat and removing phlegm method on airway inflammation in airway mucus hypersecretion rats and its mechanism. Methods: Forty-eight Sprague-Dawley rats were randomly divided into 6 groups with 8 rats in each group, which were blank group, model group, low dose Zhike Granule group, middle dose Zhike Granule group, high dose Zhike Granule group and control group, Inflammatory cells were observed by cell counting and classification. Pathological changes of lung tissue were observed by HE staining and serum levels of NE were measured by ELISA. Results: ①Cell count and classification: The neutrophils, eosinophils and lymphocytes in the blank, middle and high dose Zhike Granules group were lower than the model group and the low dose Zhike Granule group (P <0.05 or P <0.01). ② HE stain: less airway mucus was found in normal rats; inflammatory cells infiltration and secretions obstructed airway and alveolar wall of rats in the model group were broken, some of the alveoli were fused into bullae; The number of bronchial tube secretions in high-dose Zhike Granule group was less and the epithelial cells arranged neatly; the control group had fewer alveolar walls and even a few inflammatory cells. ③ The level of NE in serum NE: blank group and high-dose Zhike Granule group was lower than that in model group and Zhike Granules group (P <0.01). Conclusion: Xingbocai Zhike Granule can relieve airway inflammation in rats with airway mucus hypersecretion through the method of clearing away heat and dissipating phlegm. The mechanism may be related to the decrease of blood NE.
