目的探讨来曲唑造模多囊卵巢综合征(PCOS)大鼠血浆中血管活性肠肽(VIP)及凝血功能的变化。方法将24只SD大鼠随机分成两组(每组12只):PCOS组用来曲唑1mg/kg+1%羧基纤维素连续灌胃42d;对照组用1%羧基纤维素连续灌胃42d。第21d采血检测各组大鼠血浆中的睾酮和胰岛素,第42d采血检测VIP、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB)、血小板、睾酮和胰岛素,实验前、第21和42d称重。结果 PCOS组大鼠体重明显增加,出现高胰岛素血症、高雄血症,动情周期消失,符合多囊卵巢综合征大鼠模型;血浆中的VIP浓度低于对照组,血小板数高于对照组,PT、TT减低,FIB升高。结论来曲唑造模多囊卵巢综合征大鼠的静脉血处于高凝状态,检测VIP和凝血功能,可作为临床多囊卵巢综合征动物模型病理研究和治疗的参考指标。
Objective To investigate the changes of plasma vasoactive intestinal peptide (VIP) and coagulation function in levamisole-treated polycystic ovarian syndrome (PCOS) rats. Methods Twenty-four Sprague-Dawley rats were randomly divided into two groups (12 in each group): PCOS group was treated with letrozole 1 mg / kg and 1% carboxymethyl cellulose for 42 days. The control group was treated with 1% carboxymethylcellulose for 42 days . Blood samples were collected for the detection of testosterone and insulin in the 21st day, plasma VIP, prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT), fibrinogen FIB), platelets, testosterone and insulin, before the experiment, weighed 21 and 42d. Results The body weight of rats in PCOS group was significantly increased, with hyperinsulinemia and hypercholesterolemia. The estrous cycle disappeared, which accorded with the polycystic ovary syndrome rat model. The VIP concentration in plasma was lower than that in control group, and the platelet count was higher than that in control group. PT, TT decreased, FIB increased. Conclusions Letrozole-induced polycystic ovary syndrome rat venous blood hypercoagulable state, the detection of VIP and coagulation function, clinical polycystic ovary syndrome can be used as a clinical model of pathological studies and treatment of reference index.
