目的:观察氯化锂-毛果芸香碱慢性点燃大鼠在点燃过程中脑内腺苷激酶(ADK)表达的动态演变情况,探讨癫发作与ADK的关系及时间演变特征,以评价ADK在癫发病机制中的作用。方法:氯化锂-毛果芸香碱点燃大鼠模型,分模型组、对照组和正常组。采用RT-PCR及Western blot的方法监测发作后24h和1、6个月时大鼠脑内ADK表达的动态改变情况。结果:氯化锂-毛果芸香碱点燃后24h时,RT-PCR及Western blot检测ADK的表达均低于正常对照组,差异有统计学意义(P<0.05或P<0.01);在点燃进入慢性期后的1和6个月时则相反,ADK表达较点燃前明显增高,差异有显著统计学意义(P<0.01)。ADK表达6个月时较1个月时更高(P<0.05或P<0.01),即慢性期ADK的表达增高随时间延长越加明显。结论:ADK在点燃过程中呈现双向改变特征,急性期表达降低,促进细胞外腺苷浓度的增高;进入慢性期,反复的癫发作导致了ADK的过表达,这可能是导致癫反复发作而无法控制的重要因素。

Conclusion: ADK showed bidirectional changes in the process of ignition, decreased expression in acute phase, and increased the concentration of extracellular adenosine. Into the chronic phase, repeated epileptic seizures led to ADK overexpression, which may lead to recurrent seizures And can not control the important factors.
