目的:探讨增强核受体相关因子1(Nurr 1)表达对酪氨酸羟化酶(TH)基因修饰的骨髓间充质干细胞(MSCs)中THmRNA的表达,以及对MSCs表达TH阳性细胞数的影响。方法:TH转染组(TH组)和TH与Nurr 1共同转染组(Nurr1-TH组)转染10d后,用半定量RT-PCR检测TH mRNA的表达,细胞免疫化学方法检测TH的表达。结果:Nurr1-TH组TH mRNA表达相对水平和MSCs内TH阳性细胞百分率均高于TH组(P>0.05);空白质粒对照组未检测到TH表达。结论:TH基因修饰的MSCs能高效稳定的表达TH mRNA,Nurr 1能显著增强TH修饰的MSCs中TH mRNA的表达和TH阳性细胞数,为下一步移植治疗帕金森病奠定了基础。
OBJECTIVE: To investigate the expression of TH mRNA in bone marrow mesenchymal stem cells (MSCs) modified with tyrosine hydroxylase (TH) gene by enhancing the expression of nuclear receptor related factor 1 (Nurr 1) and the number of TH positive cells influences. Methods: The expression of TH mRNA was detected by semi-quantitative RT-PCR and the expression of TH was detected by immunocytochemical method after 10 days of transfection with TH group and Nurr1-TH group . Results: The relative expression of TH mRNA and the percentage of TH positive cells in Nurr1-TH group were higher than those in TH group (P> 0.05). No TH expression was detected in blank plasmid control group. CONCLUSION: TH gene-modified MSCs can express TH mRNA efficiently and stably. Nurr 1 can significantly enhance the TH mRNA expression and the number of TH-positive cells in TH-modified MSCs, which lays the foundation for the further transplantation of Parkinson’s disease.
