目的:研究葡萄籽提取物原花青素(procyanidins,PC)降低家兔动脉血压的作用及机制。方法:采用颈总动脉插管测量动脉血压,将动物随机分为:PC、左旋硝基精氨酸、甲烯蓝、异搏定、普萘洛尔、吲哚美辛、缬沙坦7组。PC组为静脉累积注射PC(4.0、8.0、16、32、64、84mg/kg)后,记录血压并计算下降百分率(%),其余各组先注射不同的阻断剂对血压无影响后,再注射PC(64mg/kg)测量各组血压并计算下降百分率(%)。结果:PC能显著降低家兔动脉血压,并具有量效依赖性(收缩压:r=0.92,t=17.44,P<0.01;舒张压:r=0.92,t=17.47,P<0.01)。左旋硝基精氨酸和甲烯蓝能明显减弱PC(64mg/kg)的降压作用(P<0.01);异搏定、普萘洛尔、吲哚美辛、缬沙坦对PC的降压作用无明显影响。结论:PC降低家兔动脉血压的机制可能与内皮释放的NO有关。
Objective: To study the effect and mechanism of grape seed extract procyanidin (PC) in reducing arterial blood pressure in rabbits. METHODS: Arterial blood pressure was measured using carotid artery catheterization. Animals were randomly divided into PC, L-nitro arginine, methylene blue, verapamil, propranolol, indomethacin, and valsartan 7 groups. . In the PC group, after intravenous injection of PC (4.0, 8.0, 16, 32, 64, 84 mg/kg), blood pressure was recorded and percent reduction (%) was calculated. After the other groups were injected with different blockers, there was no effect on blood pressure. Each group of blood pressure was measured by further injection of PC (64 mg/kg) and the percent reduction (%) was calculated. RESULTS: PC could significantly reduce arterial blood pressure in rabbits and was dose-effect dependent (systolic pressure: r=0.92, t=17.44, P<0.01; diastolic blood pressure: r=0.92, t=17.47, P<0.01). L-nitro-arginine and Methylene blue could significantly reduce the antihypertensive effect of PC (64mg/kg) (P<0.01), and the decrease of PC by verapamil, propranolol, indomethacin and valsartan. No significant effect of pressure. CONCLUSION: The mechanism of PC lowering arterial blood pressure in rabbits may be related to NO released from the endothelium.
