利用RT-PCR技术分析了中国水仙(Narcissus tazetta var.chinensis)花器官特征基因NT-MADS1在花期不同器官及花不同部位的表达模式。结果表明,该基因只在花器官中表达,且在雄蕊和副冠中的表达量高于雌蕊,在花瓣中未检测到该基因的表达。将该基因置于CaMV35S启动子控制下,构建到载体pBI121的多克隆位点,获得了包含35S启动子、NTMADS1基因的编码区和NOS终止区域的植物表达载体。在农杆菌介导下将该基因转入模式植物拟南芥(Arabidopsis thaliana)中,获得了卷叶、花期提早和花形改变的转基因植株。
The expression pattern of NT-MADS1, a floral organ of Narcissus tazetta var. Chinensis, was analyzed by RT-PCR in different organs and floral stages in florescence. The results showed that the gene was expressed only in floral organs, and its expression was higher in stamens and paws than in pistils. No expression of this gene was detected in petals. The gene was placed under the control of the CaMV35S promoter and constructed into the multiple cloning site of the vector pBI121 to obtain a plant expression vector containing the 35S promoter, the coding region of the NTMADS1 gene and the NOS termination region. The gene was transferred into Arabidopsis thaliana under the Agrobacterium tumefaciens transformation, and the transgenic plants with leaf roll, early flowering and change of flower shape were obtained.
