目的:研究依他酸(EA)和卡西霉素(Cal)对人胚肺成纤维细胞脱氧核糖核酸和胶原合成的影响。方法:采用[~3H]TdR与[~3H]脯氨酸掺入分别测定脱氧核糖核酸和胶原合成。结果:给药24h后,EA0.05—4mmol·L~(-1)和Cal0.25—20μmol·L~(-1)组胶原合成均显著增加,脱氧核糖核酸合成无明显改变。给药36和48h,EA 1—4mmol·L~(-1)组胶原与脱氧核糖核酸合成降低:Cal0.25—20μmol·L~(-1)组脱氧核糖核酸合成亦下降,胶原合成仅10和20μmol·L~(-1)组降低。结论:细胞外液Ca~(2+)内流促进胶原合成,细胞内Ca~(2+)浓度过高或过低均抑制脱氧核糖核酸合成。
AIM: To investigate the effects of etidronate (EA) and calicheamicin (Cal) on DNA and collagen synthesis in human embryo lung fibroblasts. Methods: [~ 3H] TdR and [~ 3H] proline incorporation were used to determine DNA and collagen synthesis, respectively. Results: The collagen synthesis of EA0.05-4mmol·L ~ (-1) and Cal0.25 ~ 20μmol·L ~ (-1) group increased significantly after 24h administration, while the DNA synthesis did not change significantly. After administration for 36 and 48h, the synthesis of collagen and DNA in EA 1-4mmol·L -1 group decreased: the synthesis of DNA in Cal0.25-20μmol·L -1 group also decreased, and the collagen synthesis was only 10 And 20μmol·L -1 group decreased. CONCLUSION: Ca ~ (2+) influx in extracellular fluid can promote the synthesis of collagen. High or low intracellular Ca ~ (2+) concentration can inhibit DNA synthesis.
