目的探讨体外培养的两株不同转移潜能的乳头状浆液性卵巢癌细胞蛋白表达差异及其意义。方法2008年1月于浙江省肿瘤医院研究所提取卵巢癌细胞株HO-8910和高转移卵巢癌细胞株HO-8910PM的细胞裂解液及培养上清液中的总蛋白,利用弱阳离子交换芯片(WCX2)和表面加强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术分析两者的差异表达蛋白峰。用CiphergenProteinChip3.2.1软件分析芯片结果。结果在细胞裂解液中检出两株细胞差异蛋白峰共11个,其中10个蛋白峰在高转移卵巢癌细胞系HO-8910PM中表达上调,1个在HO-8910PM中表达下调。在两株细胞分泌液中共检出4个差异蛋白峰,均为高转移卵巢癌细胞系表达上调。其中质荷比为3144的差异蛋白峰在两株细胞的细胞裂解液及细胞分泌液中均检出,且都为HO-8910PM表达上调。利用Swiss蛋白数据库检索分子量相匹配的蛋白质发现质荷比(m/z)3144蛋白峰可能为信号传导类蛋白CD24。结论不同转移潜能的卵巢癌细胞存在蛋白表达差异。
Objective To investigate the differences of protein expression in two papillary serous ovarian cancer cells with different metastatic potential in vitro and its significance. Methods In January 2008, the cell lysate of the ovarian cancer cell line HO-8910 and the highly metastatic ovarian cancer cell line HO-8910PM and the total protein in the culture supernatant were extracted from the Institute of Tumor Hospital of Zhejiang Province, using a weak cation exchange chip WCX2) and surface-enhanced laser desorption / ionization time-of-flight mass spectrometry (SELDI-TOF-MS) were used to analyze the differentially expressed protein peaks. Chip results were analyzed using the Ciphergen Protein 3.0.2 software. Results A total of 11 differentially expressed protein peaks were found in the cell lysate, of which 10 were up-regulated in highly metastatic ovarian cancer cell line HO-8910PM and down-regulated in HO-8910PM. Four differential protein peaks were detected in the two cell exudates, all of which were up-regulated in highly metastatic ovarian cancer cell lines. Among them, the differential protein peak with a mass-to-charge ratio of 3144 was detected in both cell lysates and cell exudates, and both of them were upregulated in HO-8910PM expression. Using the Swiss protein database to search for proteins with similar molecular weights, it was found that the mass-to-charge ratio (m / z) 3144 protein peak might be the signal transduction-like protein CD24. Conclusion The ovarian cancer cells with different metastatic potentials have different protein expression.
