为了解肝素对脂多糖 (L PS)诱导人肾小球系膜细胞 (MC)增殖和细胞因子分泌的影响 ,作者采用四甲基偶氮唑蓝比色分析法及生物活性法 ,分别测定了人肾小球 MC增殖和细胞因子的分泌情况。结果显示 :在正常培养条件下 ,人肾小球 MC分泌一定量的白细胞介素 6 (IL- 6 )和肿瘤坏死因子 α(TNF- α)。脂多糖诱导 MC增殖及分泌 IL- 6、TNF- α。高浓度的肝素 (5 0 0 U/ml)促进 MC增殖及分泌细胞因子 ,低浓度的肝素 (5 U/ml)抑制 MC增殖及分泌细胞因子。据此 ,作者认为肝素仅在低浓度抑制 MC增殖及细胞因子分泌 ,从而为临床治疗肾小球疾病和肝素剂量选择提供了一定的实验依据
To investigate the effects of heparin on the proliferation and cytokine secretion of human mesangial cells (MC) induced by lipopolysaccharide (LPS), the authors used tetramethylzirconyl blue colorimetric assay and bioactivity assay to determine Human glomerular MC proliferation and cytokine secretion. The results showed that under normal culture conditions, human glomerular mesangium secreted a certain amount of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α). Lipopolysaccharide induces MC proliferation and secretion of IL-6 and TNF-α. High concentrations of heparin (500 U / ml) promoted the proliferation and secretion of cytokines in MCs. Low concentrations of heparin (5 U / ml) inhibited MC proliferation and cytokine secretion. Accordingly, the authors believe that heparin inhibits MC proliferation and cytokine secretion only at low concentrations, providing some experimental evidence for clinical treatment of glomerular disease and heparin dose selection
