目的观察中风Ⅱ号对局灶性脑缺血大鼠脑组织核因子-κB(NF-κB))蛋白表达的影响。方法线栓法制备大鼠大脑中动脉缺血(MCAO)模型,予中风Ⅱ号口服液(10 mg·kg-1)连续灌胃1 d、3 d、7 d,并设正常对照组、模型组。Longa法进行神经功能缺损评分,TTC染色检测脑组织缺血面积,Western blot检测大鼠脑组织NF-κB p65蛋白表达。结果模型组大鼠脑组织NF-κB蛋白表达较对照组显著升高,而中风Ⅱ号能明显改善MCAO大鼠神经功能缺损评分,减少脑组织缺血面积,降低脑组织NF-κB蛋白表达。结论中风Ⅱ号可通过抑制NF-κB的过高表达,调控炎症级联反应,减轻局灶性脑缺血大鼠的炎症损伤。
Objective To observe the effect of Zhongfeng Ⅱ on the expression of nuclear factor-κB (NF-κB) in brain tissue of focal cerebral ischemia rats. Methods The middle cerebral artery occlusion (MCAO) model was established by suture method. The rats were randomly divided into three groups: normal control group, model group group. Longa method for neurological deficit score, TTC staining of ischemic area of brain tissue, Western blot detection of rat brain NF-κB p65 protein expression. Results The expression of NF-κB protein in brain tissue of rats in model group was significantly higher than that in control group. However, stroke Ⅱ could significantly improve the neurological deficit score of MCAO rats, reduce the ischemic area of brain tissue and decrease the expression of NF-κB in brain tissue. Conclusion Stroke No.2 can inhibit the inflammatory response by inhibiting the over-expression of NF-κB, regulating the inflammatory cascade.
