选择对草鱼出血病病毒(FRV)不敏感的草鱼肝组织进行细胞培养,建立了草鱼肝细胞株,命名为GLA.对GLA进行攻毒,发现其对FRV有抗性。采用电融合结合继代移核的方法,将GLA细胞(核),移植于草鱼未受精卵内,获得开口前的幼鱼8尾和存活仔鱼1尾,说明此方法能提高细胞的发育能力。通过比较体细胞工程鱼与供体、受体亲本草鱼的形态特征,发现体细胞工程鱼可数性状与供体草鱼的基本一致,初步推断体细胞工程鱼的遗传信息,是由供体细胞核提供的。
The grass carp liver tissue which was insensitive to grass carp hemorrhagic disease virus (FRV) was selected for cell culture, and a grass carp hepatocyte cell line was established and named GLA. The GLA was challenged and found to be resistant to FRV. Using electrofusion combined with subculture, GLA cells (nuclei) were transplanted into the unfertilized eggs of grass carp to obtain 8 pre-hatch larvae and 1 surviving larvae, indicating that this method can improve cell development ability. By comparing the morphological characteristics of somatic fish and donor and recipient parental carp, we found that the somatic traits of somatic fish are basically the same as that of donor grass carp.
