目的:探讨胃泌素受体拮抗剂丙谷胺(PGL)对人结肠癌裸鼠移植瘤的作用。方法:建立人结肠癌SW480细胞株裸鼠移植瘤模型,观察在PGL、5肽胃泌素(PG)作用下移植瘤的体积和重量,用放免法测CAMP、流式细胞术测DNA、蛋白质和细胞周期。结果:移植瘤的体积和重量、瘤细胞内CAMP、DNA和蛋白质含量、S和G2M期细胞数及增殖指数PG组均显著高于对照组,PGL组与对照组比均无显著差异;PG+PGL组均显著低于PG组,而与对照组比均无显著性;G0/G1期细胞数PG组明显低于对照组,PGL组与对照组比无显著差异,PG+PGL组显著高于PG组,而与对照组比无显著性。结论:PGL对人结肠癌SW480细胞株移植瘤的生长无明显影响,但可抑制PG对移植瘤的促增殖作用,其作用是通过拮抗PG促进瘤细胞内CAMP、DNA和蛋白质的合成,抑制瘤细胞从G0/G1期进入到S、G2M期
Objective: To investigate the effect of the gastrin receptor antagonist proglumide (PGL) on human colon cancer xenografts in nude mice. METHODS: A xenograft tumor model of human colon cancer SW480 cell line was established. The volume and weight of the transplanted tumor under the action of PGL and 5-peptide gastrin (PG) were measured. CAMP and flow cytometry were used to measure DNA and protein. And cell cycle. Results: The volume and weight of transplanted tumor, intratumoral CAMP, DNA and protein content, S and G2M phase cell number, and proliferation index in the PG group were significantly higher than those in the control group. There was no significant difference between the PGL group and the control group; PG+PGL group. All of them were significantly lower than those in the PG group, but they were not significantly different from the control group. The number of G0/G1 phase cells in the PG group was significantly lower than that in the control group. There was no significant difference between the PGL group and the control group. The PG+PGL group was significantly higher than the PG group. There was no significant difference from the control group. Conclusion: PGL has no significant effect on the growth of human colon cancer SW480 cell line, but it can inhibit the proliferation of transplanted tumors with PG. Its role is to inhibit the synthesis of CAMP, DNA and protein in tumor cells by antagonizing PG, and inhibit the growth of tumors. Cells enter from G0/G1 phase to S, G2M phase
