目的探讨体外培养的猪视网膜色素上皮(retinalpigmentepithelial,RPE)细胞间连接与通讯功能的关系。方法用5,6-碳氧荧光素醋酸盐进行细胞染色,光淬灭某些细胞中的荧光分子,激光扫描共聚焦显微镜观察不同连接状态的RPE细胞内的荧光恢复速率,评价缝隙连接的通讯功能。结果光漂白后的RPE细胞中的荧光强度得以恢复,与之紧密相连的细胞则有荧光下降,分别与1,2,3个细胞相连的每分钟荧光恢复速率依次是1.997±0.665,4.378±0.811,8.736±2.084。结论体外培养的猪RPE细胞缝隙连接通讯功能的强弱与细胞的连接状况有关。
OBJECTIVE: To investigate the relationship between the function of intercellular junctions and communication in cultured retinal pigment epithelium (RPE) in vitro. Methods The cells were stained with 5,6-carboxyfluorescein acetate and the fluorescence molecules in some cells were quenched by light. The rate of fluorescence recovery in different connected RPE cells was observed by laser scanning confocal microscopy. Communication function. Results The fluorescence intensity of RPE cells after photo-bleaching was restored, while the fluorescence intensity of cells closely linked to them was decreased. The rate of fluorescence recovery per minute correlated with 1,2,3 cells was 1.997 ± 0.665 , 4.378 ± 0.811, 8.736 ± 2.084. CONCLUSION: The strength of gap junctional communication function of porcine RPE cells cultured in vitro is related to the cell connection status.
