目的探讨长链非编码RNA(long non-coding RNA,LncRNA)LOC100506123对胰腺癌细胞增殖及迁移的影响。方法采用实时定量PCR(qRT-PCR)技术检测胰腺癌组织及细胞系(AsPC-1、BxPC-3、Panc-1、CFPAC-1,以正常胰腺导管上皮细胞HPDE为对照)中LOC100506123的表达情况。并通过siRNA技术下调LOC100506123表达,构建siRNA经Lipofectamine 2000~(TM)脂质体转染细胞(对照组为siR-NC,实验组为siR-1、siR-2),采用CCK-8、Transwell小室实验分别检测胰腺癌细胞增殖和转移能力。结果 1LncRNA LOC100506123在胰腺癌组织及细胞系中表达显著升高(P<0.05)。2下调LOC100506123表达,胰腺癌细胞的增殖及迁移能力显著降低(P<0.05)。结论高表达的LncRNA LOC100506123能促进胰腺癌细胞的增殖及迁移。
Objective To investigate the effect of long non-coding RNA (LncRNA) LOC100506123 on the proliferation and migration of pancreatic cancer cells. Methods The expression of LOC100506123 in pancreatic cancer tissues and cell lines (AsPC-1, BxPC-3, Panc-1, CFPAC-1 and normal pancreatic ductal epithelial cells HPDE) was detected by qRT-PCR . SiRNA was used to down-regulate the expression of LOC100506123. The siRNA was transfected into Lipofectamine 2000TM liposome (control group was siR-NC, experimental group was siR-1, siR-2) Experiments were used to detect the proliferation and metastasis of pancreatic cancer cells. Results The expression of 1LncRNA LOC100506123 in pancreatic cancer tissues and cell lines was significantly increased (P <0.05). 2 down-regulated the expression of LOC100506123, the proliferation and migration ability of pancreatic cancer cells were significantly decreased (P <0.05). Conclusion High expression of LncRNA LOC100506123 can promote the proliferation and migration of pancreatic cancer cells.
