目的:探讨人脑胶质瘤组织中两种耐药基因ERCC2、MGMT表达之间的相关性及其与预后的关系。方法:采用双管RT-PCR方法检测19例人脑胶质瘤组织中ERCC2和MGMTmRNA水平,并分析它的相关性,分析其与患者生存时间的关系。结果:ERCC2、MGMT的表达量分别为1.42±0.93、2.89±1.82,ERCC2与MGMT之间具有极显著的相关性(p<0.01)。与患者的生存时间具有显著的相关性(p<0.05)。结论:ERCC2、MGMT基因表达之间具有内在的联系,共同参与细胞对DNA损伤的修复,并形成肿瘤耐药;与肿瘤的预后具有明显的相关性。
Objective: To investigate the correlation between the expression of ERCC2 and MGMT in human glioma tissue and its relationship with prognosis. Methods: The mRNA and protein levels of ERCC2 and MGMT in 19 human glioma tissues were detected by double-tube RT-PCR. The correlation between ERCC2 and MGMT mRNA levels was analyzed. The relationship between ERCC2 and MGMT mRNA expression and survival time was analyzed. Results: The expressions of ERCC2 and MGMT were 1.42 ± 0.93 and 2.89 ± 1.82, respectively. There was a significant correlation between ERCC2 and MGMT (p <0.01). There was a significant correlation with patient’s survival time (p <0.05). CONCLUSION: The expression of ERCC2 and MGMT genes are intrinsically linked and participate in the repair of DNA damage and the formation of drug resistance in cancer cells. The expression of ERCC2 and MGMT genes has a significant correlation with the prognosis of cancer.
