目的:研究螺旋藻硒多糖(polysaccharide from Se-enriched spirulina platensis,Se-PSP)各组分对肿瘤细胞生长的影响。方法:超声波法从富硒螺旋藻(Se-enriched spirulina,Se-SP)中提取Se-PSP总糖和各组分(Se-PSP1、Se-PSP2、Se-PSP3和Se-PSP4),DEAE-纤维素层析对各组分进行分离纯化。MTT法检测10μg/ml时Se-PSP总糖及不同组分对卵巢癌SKOV-3细胞生长的影响;观察不同质量浓度的Se-PSP2对卵巢癌SKOV-3细胞、肝癌HepG-2细胞、胃癌BGC-803细胞、鼻咽癌CNE细胞和舌鳞癌Tca-8113细胞生长的影响。结果:Se-SP分离纯化得到4个不同组分(Se-PSP1、Se-PSP2、Se-PSP3和Se-PSP4),其中Se-PSP2质量分数最高。10μg/mlSe-PSP总糖及其4个组分对SKOV-3细胞生长的抑制率分别为(33.393±5.437)%、(22.290±8.399)%、(44.881±4.878)%、(45.354±4.931)%和(46.203±4.455)%,其中Se-PSP2、Se-PSP3和Se-PSP4抑制率显著大于Se-PSP总糖及Se-PSP1(P<0.05),但前3者无明显差别(P>0.05)。Se-PSP2能剂量依赖性抑制SKOV-3、HepG-2、BGC-803和Tca-8113肿瘤细胞的增殖(均P<0.05),抑制作用具有肿瘤广谱性。结论:Se-PSP2是Se-PSP的主要成分,对多种肿瘤细胞的生长有抑制作用,有望成为治疗肿瘤的辅助药物。
Objective: To study the effect of polysaccharide from Se-enriched spirulina platensis (Se-PSP) on tumor cell growth. Methods: The total sugar and components of Se-PSP (Se-PSP1, Se-PSP2, Se-PSP3 and Se-PSP4) were extracted from Se-enriched spirulina (Se- The constituents were separated and purified by cellulose chromatography. The effects of different concentrations of Se-PSP2 on the proliferation of SKOV-3 ovarian cancer cells, HepG-2 hepatocellular carcinoma cells, gastric cancer BGC-803 cells, nasopharyngeal carcinoma CNE cells and tongue squamous cell carcinoma Tca-8113 cells. RESULTS: Four different components (Se-PSP1, Se-PSP2, Se-PSP3 and Se-PSP4) were isolated and purified from Se-SP. Se-PSP2 was the highest. The inhibitory rates of total 10μg / ml Se-PSP and its 4 components on SKOV-3 cells were (33.393 ± 5.437)%, (22.290 ± 8.399)%, (44.881 ± 4.878)% and (45.354 ± 4.931) % And (46.203 ± 4.455)% respectively. The inhibition rates of Se-PSP2, Se-PSP3 and Se-PSP4 were significantly higher than those of Se-PSP and Se- PSP1 (P < 0.05). Se-PSP2 could inhibit the proliferation of SKOV-3, HepG-2, BGC-803 and Tca-8113 tumor cells in a dose-dependent manner (all P <0.05). Conclusion: Se-PSP2 is the main component of Se-PSP, which can inhibit the growth of many tumor cells and is expected to become an adjuvant drug for tumor treatment.
