目的:研究抗氧化微量营养素(AM)对1型糖尿病鼠胰岛细胞的保护作用。方法:应用小剂量多次注射链脲佐菌素 (MLDS)的 方法制备IDDM 模型;将雄性C57BL小鼠随机分成 MLDS 模型组( MLDS )、联合添加微量营养素组(AM)、及正常对照组(Control);联合添加AM包括硒(Se)、维生素E(VE)、钒(V)、铬(Cr),给药7 周后处死动物。制备小鼠胰腺HE切片,测定胰岛面积与胰腺面积比 (Isl.A/Panc.A)、胰岛平均面积 (Isl.A)、胰岛形状因子( IFF ); 并应用流式细胞术(FCM)测定外周血淋巴细胞TNF-α、IFN-γ、IL-4、IL-10抗原表达。结果:MLDS模型组Isl.A/Panc.A 、Isl.A明显低于AM组和正常对照组(p<0.05或p<0.01); IFF 显著高于AM组和正常对照组(p<0.05或p<0.01)。模型组小鼠外周血淋巴细胞TNF-α抗原表达明显高于AM组和正常对照组(p<0.01),IFN-γ表达高于正常对照组(p<0.05),IL-10表达显著低于AM组和正常对照组(p<0.01)。结论:AM通过抗氧化及免疫调节作用可减轻致胰岛损伤因素引起的胰岛炎、胰岛内分泌细胞变性及萎缩改变,在维持胰岛正常结构与改善胰岛功能方面具有一定的作用。
Objective: To study the protective effects of antioxidant micronutrients (AMs) on islet cells in type 1 diabetic mice. Methods: The IDDM model was established by injecting streptozotocin (MLDS) in small doses. The male C57BL mice were randomly divided into three groups: MLDS, ML group, AM group and normal control group (AM) including selenium (Se), vitamin E (VE), vanadium (V) and chromium (Cr) The pancreatic sections of mice were prepared and the ratio of islet area to islet area (Isl.A / Panc.A), the average islet area (Isl.A) and islet shape factor (IFF) were measured. Flow cytometry (FCM) The expression of TNF-α, IFN-γ, IL-4 and IL-10 antigen in peripheral blood lymphocytes. Results: The levels of Isl.A / Panc.A and Isl.A in MLDS model group were significantly lower than those in AM group and normal control group (p <0.05 or p <0.01), respectively. IFF was significantly higher than that in AM group and normal control group (p <0.05 or p <0.01). The expression of TNF-α in peripheral blood lymphocytes in model group was significantly higher than that in AM group and normal control group (p <0.01), and the expression of IFN-γ was higher than that in normal control group (p <0.05) AM group and normal control group (p <0.01). Conclusion: Antioxidant and immunomodulatory effects of AM can reduce insulitis, islet endocrine cell degeneration and atrophy induced by pancreatic islet injury, and play an important role in maintaining normal pancreatic islet structure and improving pancreatic islet function.
