目的探讨白芍总苷脂质体对人胃癌BGC-823细胞凋亡的诱导作用。方法 CCK-8试剂盒检测白芍总苷脂质体对BGC-823细胞增殖的影响,HE染色、Hochest 33342/PI双染和透射电子显微镜观察BGC-823细胞凋亡的形态学变化,Western blotting检测细胞凋亡相关因子Bcl-2和Bax蛋白的表达情况。结果白芍总苷脂质体能明显抑制BGC-823细胞的增殖,呈现一定的剂量依赖关系(P<0.05)。白芍总苷脂质体干预BGC-823细胞48 h后,凋亡形态学观察均可见细胞核固缩、碎裂和凋亡小体等凋亡特征改变,Western blotting检测Bcl-2蛋白表达减少,Bax表达增多,Bcl-2/Bax比值降低。结论白芍总苷脂质体明显诱导胃癌BGC-823细胞凋亡,抑制其增殖,其机制可能与下调Bcl-2蛋白、上调Bax蛋白及降低Bcl-2/Bax比值有关。
Objective To investigate the effects of total glucosides of peony on the apoptosis of human gastric cancer BGC-823 cells. Methods CCK-8 kit was used to detect the effect of total glucosides of peony on the proliferation of BGC-823 cells. HE staining, Hochest 33342 / PI double staining and transmission electron microscopy were used to observe the morphological changes of BGC-823 cells. Western blotting The expression of apoptosis related factors Bcl-2 and Bax protein were detected. Results Total saponin liposomes can significantly inhibit the proliferation of BGC-823 cells in a dose-dependent manner (P <0.05). The apoptosis of BGC-823 cells treated with total peptidoglycan liposomes for 48 h showed that apoptotic features such as nuclear condensation, fragmentation and apoptotic bodies were observed. The expression of Bcl-2 protein was decreased by Western blotting, Bax expression increased, Bcl-2 / Bax ratio decreased. Conclusion The total glucosides of peony root can obviously induce the apoptosis of gastric cancer cell line BGC-823 and inhibit its proliferation. The mechanism may be related to down-regulating Bcl-2 protein, up-regulating Bax protein and decreasing Bcl-2 / Bax ratio.
