本文报告用扫描电镜观察体外培养的人胎儿心肌细胞发育的形态特征。材料来自妊娠10、13、16周人胎儿心脏。首先进行组织块培养,然后观察由培养团块上新生并脱离团块贴璧生长的心肌细胞。培养早期(脱落贴壁生长1周左右),细胞呈球形、短梭形,表面有各种突起。中期(脱落生长约2~4周)细胞呈扁平不规则形。细胞间由突起相连接。晚期(脱落生长超过1个月)部分细胞可发育成带状、柱状,由闰盘样结构连接成分支状。体外培养的胎儿心肌细胞,在发育过程中,可见闰盘样结构形成的过程,通过这种连接,使一群细胞相互连接,平行排列成束,近似于成熟心肌细胞的形态及排列。本文研究提示,体外培养的人胎儿心肌细胞似有分化、发育成熟的趋向。
This article reports the morphological characteristics of human fetal cardiomyocytes cultured in vitro using scanning electron microscopy. Materials from human fetal heart rate at 10, 13, 16 weeks of gestation. First, tissue culture, and then observed by the training of new clumps on and off the clumps of growth of cardiomyocytes. Early culture (off adherent growth for about 1 week), the cells were spherical, short fusiform, the surface of a variety of processes. In the interim (about 2 to 4 weeks of exfoliation) the cells showed a flat, irregular shape. Cells are connected by protrusions. Late (shedding more than 1 month) part of the cells can develop into a banded, columnar, connected by the intercalated disc-like structure of the branch. In vitro culture of fetal cardiomyocytes, in the development process, we can see intercalated disc-like structure of the formation process, through this connection, so that a group of cells connected to each other, arranged in parallel into a bundle, similar to mature cardiomyocytes morphology and arrangement. This study suggests that human fetal cardiomyocytes cultured in vitro seem to be differentiated and mature.
