目的通过观察肝豆灵片对铜负荷大鼠肝细胞凋亡及肝组织核转录因子kappa B(NF-κB)和半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)表达的影响,分析肝豆灵片对铜负荷大鼠肝损伤治疗的作用机制。方法18只Wistar大鼠被随机分为对照组、模型组和肝豆灵组,每组6只。对照组自由饮食,模型组和肝豆灵组连续12周以铜负荷法复制大鼠模型,自第7周开始,肝豆灵组大鼠以肝豆灵片混悬液灌胃,直至造模结束。采用HE染色法观察大鼠肝组织病理变化,TUNEL法检测大鼠肝细胞凋亡,免疫组化技术检测大鼠肝组织NF-κB和Caspase-3的表达。结果与对照组比较,模型组大鼠肝组织病理损伤严重,肝细胞凋亡指数显著升高(P<0.05),NF-κB和Caspase-3表达显著升高(P<0.05);与模型组比较,肝豆灵组大鼠肝组织病理损伤减轻,肝细胞凋亡指数及NF-κB,Caspase-3表达显著降低(P<0.05)。结论肝豆灵片可减轻铜负荷大鼠肝损伤,其机制可能与其下调肝组织NF-κB和Caspase-3的表达、抑制肝细胞凋亡相关。
Objective To observe the effects of GGT on the apoptosis of hepatic cells and the expression of nuclear factor kappa B (NF-κB) and Caspase-3 in rats with copper overload, The mechanism of action of Gudou Ling tablet in the treatment of liver injury induced by copper in rats was analyzed. Methods Eighteen Wistar rats were randomly divided into control group, model group and liver bean group, with 6 rats in each group. Control group of free diet, model group and liver bean group for 12 weeks by copper load replication rat model, starting from the 7th week, liver bean group rats with Gudouling tablets suspension gavage until modeling End. The pathological changes of liver were observed by HE staining. The apoptosis of liver cells was detected by TUNEL method. The expressions of NF-κB and Caspase-3 were detected by immunohistochemistry. Results Compared with the control group, the pathological changes of hepatic tissue in the model group were severe and the hepatocyte apoptosis index (P <0.05) and the expression of NF-κB and Caspase-3 were significantly increased (P <0.05) Compared with the control group, the hepatic tissue injury of liver was alleviated and the hepatocyte apoptotic index and the expressions of NF-κB and Caspase-3 were significantly decreased (P <0.05). Conclusion GGT can reduce hepatic injury induced by copper in rats, and its mechanism may be related to down-regulation of NF-κB and Caspase-3 in liver tissue and inhibition of hepatocyte apoptosis.
