利用来自小麦、大麦、玉米、高粱、水稻的1058对EST-SSR在翦股颖上进行扩增,得到有效扩增引物620对(58.60%)。其中源于小麦、大麦、玉米、高粱、水稻的引物对数分别为313(61.49%)、30(63.83%)、101(51.53%)、11(35.48%)、165(60.00%)。进一步利用其中53个引物对,对8个翦股颖品种进行了遗传多样性分析。有51对引物显示了多态性,占引物数的94.34%;共检测到368个等位变异,平均每对引物有6.9个等位变异;多态性信息指数(PIC)为0.49~0.93,平均为0.78;品种间遗传相似系数为0.48~0.86,平均为0.72。聚类分析结果表明,在相似系数0.75处,8个品种可明确的分为两大类,第Ⅰ类为高地;第Ⅱ类又可分为两个亚类,Ⅱ-1亚类由阿尔法、潘克劳斯、回旋组成,Ⅱ-2亚类由普特、L-93、开拓、A4组成。
1058 pairs of EST-SSRs from wheat, barley, maize, sorghum and rice were amplified on Agrobacterium tumefaciens to obtain 620 pairs of effective amplification primers (58.60%). Among them, the number of primers derived from wheat, barley, maize, sorghum and rice was 313 (61.49%), 30 (63.83%), 101 (51.53%), 11 (35.48%) and 165 (60.00%), respectively. Further analysis of the genetic diversity of 8 wild plants by using 53 primer pairs. 51 pairs of primers showed polymorphism, accounting for 94.34% of the total number of primers. A total of 368 alleles were detected with an average of 6.9 alleles per primer pair. The polymorphism information index (PIC) ranged from 0.49 to 0.93, With an average of 0.78. The genetic similarity coefficient was 0.48 ~ 0.86, with an average of 0.72. The results of cluster analysis showed that at the similarity coefficient of 0.75, eight cultivars could be clearly divided into two categories, the first is highland, the second is divided into two subtypes, and the second subunit is composed of alpha, Panklaus, composed of cyclotron, II-2 sub-category by Put, L-93, pioneering, A4 composition.
