目的研究发现早期高压氧(HBO)治疗可以减轻缺氧缺血性脑损伤(HIBD)新生大鼠的脑白质损伤。但延迟开始治疗的时间是否也有同样的作用尚不清楚。该文探讨不同时间窗HBO治疗对HIBD新生大鼠脑白质损伤的影响,从而探讨HBO治疗HIBD的时间窗。方法采用Rice法制成HIBD模型,分别于HIBD后3,6,12,24,72h开始给予HBO治疗,每日1次,连续7d。HIBD后14天(生后21天)开始采用盲法进行T迷宫试验、放射性迷宫测试、足错误测试行为学测试;测试完毕,用髓鞘碱性蛋白(MBP)免疫组化法检测胼胝体与纹状体区髓鞘蛋白。结果HIBD后3,6,12hHBO治疗各组T迷宫测试、放射性迷宫试验和足错误测试的测试结果均优于HIBD模型组(P<0.05)。HIBD后24,72hHBO治疗组行为学测试结果与HIBD模型组比较,差异无显著性意义。HIBD后3,6,12hHBO治疗各组大脑皮层胼胝体与纹状体区域MBP吸光度显著高于HIBD模型组,差异有显著性意义(P<0.05),而HIBD后24,72hHBO治疗组与HIBD模型组相比差异无显著性。结论HIBD后12h内HBO治疗可以显著改善脑白质损伤,HIBD后24h治疗则疗效不显著。
Objective To study the effects of early hyperbaric oxygen therapy (HBO) on the white matter damage in neonatal rats with hypoxic-ischemic brain damage (HIBD). However, it is unclear whether the delay in starting treatment has the same effect or not. This article explored the effect of HBO treatment at different time windows on white matter damage in neonatal rats with HIBD and then explored the time window of HBO treatment for HIBD. Methods The HIBD model was made by the method of Rice. HBO treatment was given at 3, 6, 12, 24 and 72 h after HIBD, once daily for 7 days. After 14 days of HIBD (21 days after birth), T-maze test, radioactive maze test and foot erroneous test were performed blindly. After the test, MBP immunohistochemistry was used to detect the relationship between the corpus callosum and Myometrium myelin protein. Results The results of T maze test, radioactive maze test and foot mistaken test in HH group were better than HIBD model group (P <0.05). HIBD after 24,72hHBO treatment group behavioral test results compared with the HIBD model group, the difference was not statistically significant. The absorbance of MBP in corpus callosum and striatum of HIBD treated groups was significantly higher than that of HIBD model group at 3, 6 and 12h after HIBD treatment (P <0.05), while HIBD treatment group and HIBD model group No significant difference compared. Conclusions HBO treatment can significantly improve the white matter damage within 12 hours after HIBD, but not after 24h HIBD treatment.
